Riyaz Khan is a PhD researcher in the Laboratory for Systems and Synthesis Immunology (LSSI), led by Prof. Sai Reddy, at ETH Zürich's Department of Biosystems Science and Engineering (D-BSSE) in Basel. His work focuses on engineering allogeneic T-cell therapies, bringing together T-cell receptor engineering, bispecific antibody synergy, and immune-evasion strategies to move cell therapy toward safe, off-the-shelf products. His research spans both wet-lab immunology and computational protein design. He is an inventor of the T-PRO allogeneic T-cell platform, now being advanced toward clinical translation through Encelta AG.
This talk will explore the use of engineered allogeneic T cells in enhancing bispecific antibody (biAb) cancer immunotherapy. By decoupling T-cell receptor (TCR) binding from CD3 signaling, we maintain biAb-driven T-cell activation while preventing harmful alloreactive responses. In a CD19+ tumor model, these engineered T cells, combined with the biAb blinatumomab, demonstrated effective tumor clearance without detectable alloreactivity, highlighting a promising approach for ‘off-the-shelf’ T-cell therapies.
