Olga Del Perugia is a Research Associate Scientist at Philochem AG, Switzerland, and a Doctoral student at the University of Pavia (IUSS), Italy, specializing in cell line development. She holds a degree in Neuroscience from the University of Pisa and a bachelor’s degree in Biological Sciences from the University of Florence. Her doctoral work focuses on developing a site-specific integration (SSI) platform to generate stable producer cell lines for recombinant protein manufacturing.
Antibodies are pivotal in immunotherapy, but cost-effective GMP production depends on robust, high-yield expression systems. Traditional random transgene integration (RTI) often yields unpredictable expression, while site-specific integration (SSI) ensures molecularly defined, reproducible insertion events, reducing clonal variability and accelerating the development of reliable producer clones. In this talk, I will present a hybrid CRISPR/Cas9 and recombinase-mediated cassette exchange approach to generate stable cell lines for recombinant protein expression. I will discuss results comparing productivity between RTI and SSI clones across genomic loci and how this platform enables fine-tuning of expression cassettes for challenging proteins.
