Genovis provides unique enzymes and technologies for rapid characterization and conjugation of antibody-based molecules, e.g. mAbs, ADCs, Fc-fusion proteins, and biosimilars. The new GlyCLICK™ conjugation technology allows site-specific labeling of functional groups at the Fc-glycans of IgG. The technology consists of two steps enzymatic remodeling of the Fc-glycan and a click chemistry conjugation. The site-specificity at the Fc ensures an intact antigen binding capacity of the antibody. Applications of the GlyCLICK technology includes not only ADC and mAb development, but also microscopy and in vivo imaging. The SmartEnzymes™ portfolio consists of FabRICATOR® (IdeS), FabRICATOR®Z (IdeZ), FabULOUS® (SpeB), GingisKHAN® (KGP), GingisREX® (Rgp), IgGZERO® (EndoS) and GlycINATOR® (EndoS2). The specific proteases are applied in characterization of antibody based therapeutics often using liquid chromatography and mass spectrometry. The FabRICATOR enzyme specifically and rapidly cuts antibodies below the hinge, and after reduction, three 25 kDa subunits can be studied with increased resolution. The applications include monitoring of oxidation on antibodies, glycosylation studies, identity testing, comparability studies of biosimilars, and many others. GingisKHAN digests IgG1 above the hinge, generating intact and homogenous Fab fragments in one step. This enzyme can be used to study bispecific antibodies or to generate Fab fragments for determination of monovalent binding affinities.
