My position is as a physician-scienstist doing both lab-based research in stem cell biology and clinical work in hematology/BMT and cell-based immunotherapies.
Studies in the Kaufman Lab focus on use of human pluripotent stem cells to study basic mechanisms that regulate early human blood cell development and to derive therapeutic mature blood cell populations. These studies utilize both human embryonic stem cells (hESCs) and human induced pluripotent stem cells (iPSCs). hESCs and iPSCs provide an ideal platform both for studies of human hematopoiesis and large-scale production of blood cells such as lymphocytes that can be used for novel therapies against cancer and other diseases. We are now pursuing several strategies to move hESC/iPSC-derived NK cells into clinical therapies. This includes 1) Large scale production of hESC/iPSCs-NK cells to treat hematological malignancies (blood cell cancers), 2) engineering iPSC-NK cells with chimeric antigen receptors (CARs) to target refractory tumors to produce a standardized, off-the-shelf, targeted immunotherapy against both relapsed/refractory solid tumors and hematological malignancies, and 3) Production of hESC/iPSC NK cells with improved expression of CD16 to mediate more effective antibody-dependent cell-mediated cytotoxicity (ADCC) and more potent anti-tumor activity. We are also working on more efficient development of other blood cell populations including monocytes/macrophages and T cells that can also be used therapeutically. I also serve as Director of the Cell Therapy program at UCSD with the goal to bring more cell-based therapies to clinical trials for malignant and non-malignant diseases. I have trained many graduate students and post-doctoral fellows. In addition to NIH and CIRM funded research and research support form private foundations and industry collaborators, I previously founded and was PI on an NIH T32 Training Grant in “Stem Cell Biology”
